In today’s world, biomanufacturing has paced up synthetic biology in the realm of biological engineering. Our lab at NCCS is engineering novel signaling pathways in order to scale up the signaling prototype. A synthetic gene circuit with a positive feedback loop that activates a signaling network component during infection can be integrated with a reconstructed signaling model. With the addition of a feedback loop mechanism, a wide range of repressors, promoters, activators, and enzymes can be viewed as modules for creating a synthetic circuit that targets the immunological signal response. This might make it possible to stop immune-evading techniques and make a biomaterial that can be used in biomedical applications. Additionally, our lab at NCCS wants to induce coordinated biological activity in a group of immune cells by means of intercellular communication. Using a relatively simple genetic circuit and a synthetic biology approach, we are able to understand the dynamics of signaling and assign functions to network motifs. If certain conditions are met, such as sufficient amplification of the signal and appropriate promoter strength matching, the signal can spread throughout a cell population. Therefore, introducing a synthetic gene circuit that is connected to a biological activity like macrophage immunological signaling and has a positive feedback loop could precisely fine-tune transgene expression in a diseased state. An output from one process could be used as an input signal in the design of a gene circuit to create artificial cell-cell communication that is tunable. Using a synthetic biology approach, this artificial cellular circuitry behavior response can be seen and shown experimentally in mammalian cells. Inspired by the natural regulatory mechanism in infected human macrophages, this artificial synthetic signaling system may prove to be a flexible tool for controlling the expression of genes through gene regulatory networks. Using macrophages as a potential therapeutic intervention system is an appealing idea since they are extremely adaptable and flexible immune system cells that are responsive to activation by direct sensing of pathogen during an infection.

Bioinformatics, Computational Structural Biology and Systems Biology
The lab focuses on molecular dynamics simulation of proteins and their interacting molecules. We have shown structural information about drug-protein complexes fundamental for drug discovery via virtual screening and how it guides the optimization of the pharmacological properties of these molecules. Lately, we have started working on systems biology where we aim to integrate the action of regulatory circuits, cross-talk between pathways and the non-linear kinetics of biochemical processes through mathematical models. The current thrust is to explore the possibility of network-based drug design and how rationalized therapies may benefit from Systems Biology.
Drug Delivery and Translational Research
The lab is focused on topics such as designing and developing novel drug delivery systems, nanomedicine; with a focus on their application to disease conditions; short-term and long-term biocompatibility of drug delivery systems, host response and devices for drug delivery and drug/device combination products.
